There used to be little to no signs on that route but that is getting better by the year. The roads have been constantly fixed and widened, so it is a much more hassle free drive than it used to be a few years back when night driving was an extreme sport. I killed a dog and wrecked a car in two separate night accidents on that route and I am accident free in the rest of the world (which includes driving on ALL continents except of Africa).
So, if you have never driven in Ecuador, my first recommendation still is to arrive before nightfall. If you want to take the scenic route, follow the route along the coast (Ruta del Sol). It will take you maybe 4 hours. You will find the part Guayaquil – Salinas very well maintained, painted and with signs and everything.
If you want to be faster, but not so scenic, go directly from Guayaquil to Jipijapa (about 2 hours drive to that point). From Jipijapa you should be in Crucita in another hour. We got stranded in Crucita once and a local offered us a bed in his house over night, so we have good memories.
Mileage is secondary in Guayaquil, since road quality varies. I used to say that Manabi was one huge pothole you hit the moment you left Guayas, but it has gotten better under Correa. Still, you are more interested in the actual travel time than mileage.
Sign quality varies as well (but there are signs) depending on the road stretch, but take a map and you should be fine as long as there is light out. Beware of the “policias muertos”, the speed bumps on the main routes at high speeds, not all of them are clearly marked, especially in Manabi, which sometimes makes you see them too late especially at night (my car can flyyy).
